性质:蛋白质溶菌酶是由129个氨基酸组成的均一链的多肽。为白色冷冻干粉或结晶。等电点为10.5-11.0。酸性溶液较为稳定,加热到55℃无影响,能溶于水。

制备方法:从蛋清中提取制得。1.原料处理 新鲜或冰冻蛋清(或蛋壳水)70kg,用试纸测其pH值,应在8.0左右,经解冻后通过铅筛,除去蛋清中的脐带、蛋壳碎片及其他杂质。2.吸附 用冰水冷却并搅拌蛋清,使温度降到5℃,缓缓加入724树脂10kg,搅拌,使树脂完全悬浮在蛋清中。保持温度0-10℃,再继续搅拌5h。将蛋清移入冷库,在0-10℃静置过液。3.洗涤、洗脱 隔天把上层蛋清清液倾出,下层用清水洗去附着的蛋白质,先后共洗四次。最后将树脂抽滤去水分,用24kg0.15M磷酸缓冲液(pH6.5)分三次加入树脂搅拌,每次搅拌15min后抽滤去水分。然后用10%硫酸铵18kg,分4次洗脱溶菌酶。合并洗脱液,加入32%(重量/体积)固体硫酸铵使含量达40%,析出白色沉淀。在冷处放置过夜,过滤出沉淀。4.透析 将沉淀溶解于1.5L蒸馏水中,在冷库内透析24-36h,除去大部分硫酸铵。5.盐析 透析完毕,将溶液用滤纸过滤,得澄清透析液。用滴管慢慢加入氢氧化钠溶液,使pH上升到8.5-9.0,如有白色沉淀,应即离心除去。然后在搅拌下加入3N盐酸,使溶液pH为3.5。按体积缓缓加入5%固体氯化钠,生成白色沉淀。在0-10℃冷库放置48h,抽滤得到溶菌酶沉淀。6.干燥沉淀内加入10倍量0℃无水丙酮,搅拌使颗粒松细,在冷处静置数小时,滤去丙酮。沉淀用五氧化二磷真空干燥,即得口服或外用溶菌酶粉剂原料。如不用丙酮脱水,可将溶菌酶透析液冷冻干燥可得不含氯化钠的溶菌酶成品。按蛋清重量计,收率0.1%。

用途:主要用于五管科多种粘膜疾患,如副鼻窦炎、慢性鼻炎、咽喉炎、扁平苔癣、口腔溃疡、渗出性中耳炎等徉患,亦用于皮肤科带状包疹,扁平疣及铬性鼻炎。
