性质:又称DNA解旋蛋白。系解链酶或解链蛋白类之一。至今在大肠杆菌中发现的DNA螺旋酶就有数种,分别称为螺旋酶Ⅰ、Ⅱ、III和rep蛋白等,都与大肠杆菌DNA复制有关。作用是催化双螺旋DNA的解旋和解链,在这一解旋的过程中需水解ATP提供能量(切开一个A-T对约需5kJ·mol-1,切开一个C-G对约需21kJ·mol-1)。在复制过程中,螺旋酶Ⅱ、III沿着随从链的模板,从5′→3′方向移动,促使复制叉向前延伸;而rep蛋白则是沿着前导链的模板,以3′→5′方向向前移动,从而促进DNA双链不断地解开,让引物酶、DNA聚合酶、DNA连接酶等可有序地发挥作用,以完成DNA的复制过程。
