Objective: To establish a nested reverse transcription-polymerase chain reaction (nested RT-PCR) assay for detectioning specific fragment of human Rhinovirus (HRV) RNA,and to analyze the sequence of amplicons.
研究目的: 建立套式逆转录聚合酶链反应(nested reverse transcription-polymerase chain reaction,nested RT-PCR)检测临床样本鼻病毒(human rhinovirus,HRV)基因的方法,并进一步对扩增产物进行序列分析;用套式RT-PCR及直接免疫荧光(direct immunofluorescence assay,DFA)的方法对包括HRV在内的引起小儿急性下呼吸道感染(acute low respiratory tract infection,ALRTI)的常见病毒病原进行检测,以了解上海地区ALRTI患儿病毒感染的现状。

For each sample,RNA was extracted using commercial kit and7Nest RT-PCR using a14-pair different SARS-associated coronavirus(SARS-CoV)special primers were carried out simultaneously.
方法以2003年5月16日至23日在本院住院的46例临床确诊SARS患者为研究对象,连续留取粪便标本共103份,提取RNA后采用14对SARS-CoV特异性引物,对每份标本同时进行7次套式RT-PCR扩增,只要有1次RT-PCR结果阳性即判定该标本为阳性。
