本研究参照GenBank收录的绵羊凝乳酶原前体cDNA序列(GenBank No:X53037)设计RT-PCR、PCR与巢式引物,以新生5天的西农萨能羊公羔羊为研究对象,利用手术法采集羔羊皱胃组织,利用Trizol法提取皱胃组织总RNA,经过电泳鉴定后,以总RNA为模板利用RT-PCR方法获得西农萨能羊凝乳酶原前体cDNA序列,以cDNA序列为模板进行扩增和T载体连接测序。
