性质:破碎细胞的一种方法。将细胞在低温下突然冷冻(-15℃)和室温下融化,反复多次而达到破壁作用。由于冷冻,既能使细胞膜的疏水键结构破裂,从而增加了细胞的亲水性能,又因胞内水结晶,使细胞内外溶液浓度变化,引起细胞突然膨胀而破裂,本法适用于细胞壁较脆弱的菌体。
