性质:大肠杆菌DNA聚合酶I是一种依赖于DNA的DNA聚合酶,分子量109kD。它有5′→3′DNA聚合酶活性,5′→3′及3′→5′外切酶活性。经蛋白酶(现在是利用枯草杆菌蛋白酶)作用后得到的一大片段肽键(分子量76kD),切去了5′→3′的外切酶活性,另二酶不受影响。在基因工程技术中常用于补平限制酶切割DNA后产生的3′凹端,进行末端标记;合成cDNA第二链;在体外诱变中从单链模板中合成双链DNA;用于双脱氧末端终止法进行DNA测序;以及也可用于PCR。
