In this work, we reported a method for the purification of pepsinogens from human gastric mucosa with high pressure gel filtration chromatography(HPLC) and medium pressure anion exchange chromatography(MPLC).

【性状】
本品为白色或淡黄色的粉末；无霉败臭；有引湿性；水溶液显酸性反应。
【鉴别】
取本品的水溶液，加鞣酸、没食子酸或多数重金属盐的溶液，即发生沉 淀。
【检查】
干燥失重 取本品，在100 ℃干燥4 小时，减失重量不得过5.0 ％（附 录Ⅷ L）。
【效价测定】
对照品溶液的制备 精密称取经 105℃干燥至恒重的酪氨酸 适量，加盐酸溶液〔取1mol/L盐酸溶液65ml，加水至1000ml 〕制成每1ml 中含0.5mg 的溶液。 供试品溶液的制备 取本品适量，精密称定，用上述盐酸溶液制成每1ml 中约含 0.2 ～0.4单位的溶液。 测定法 取试管6 支，其中3 支各精密加入对照品溶液1ml ，另3 支各精密加入供 试品溶液1ml ，置37℃±0.5℃水浴中，保温5 分钟，精密加入预热至37℃±0.5℃的血红 蛋白试液5ml ，摇匀，并准确计时，在37℃±0.5℃水浴中反应10分钟。立即精密加入5% 三氯醋酸溶液5ml ，摇匀，滤过，取续滤液备用。另取试管2 支，各精密加入血红蛋白试液 5ml ，置37℃±0.5℃水浴中保湿10分钟，再精密加入 5％三氯醋酸溶液5ml ，其中1支加供 试品溶液1ml ，另1 支加上述盐酸溶液1ml ，摇匀，滤过，取续滤液，分别作为供试品和对 照品的空白对照，照分光光度法（附录Ⅳ A）在275nm 的波长处测定吸收度，算出平均值ＡS和Ａ， 按下式计算。 Ａ×Ｗｓ×ｎ 每1g含蛋白酶活力(单位)＝───────────── Ａs ×Ｗ×10×181.19 式中 ＡS 为对照品的平均吸收度； Ａ为供试品的平均吸收度； Ｗs 为对照品溶液每1ml中含酪氨酸的量，μg； Ｗ为供试品取样量，g； ｎ为供试品稀释倍数。 在上述条件下，每分钟能催化水解血红蛋白生成1μmol 酪氨酸的酶量，为一个蛋 白酶活力单位。
