性质:无色、无臭的单斜结晶。熔点98℃。几乎不溶于水,可溶于一般有机溶剂。味先苦而后麻。其盐酸盐,C17H21NO4·HCI,无色结晶或白色结晶性粉末,熔点197℃以上。极易溶于水,易溶于乙醇,溶于氯仿,不溶于乙醚。

制备方法:古柯碱是一种莨菪烷型生物碱,可由植物古柯叶中提取。古柯叶粉用乙醇提取,提取液回收乙醇、减压浓缩,浓缩液用氯仿提取,得总生物碱。古柯碱为酯类,用酸或碱水解时,生成苯甲酸、甲醇、L-爱康宁,此反应可逆向进行,将爱康宁酯化、酰化,能使其中所有爱康宁的衍生物转变为古柯碱。将古柯碱和丙酮一起加热至40℃,溶解后加入活性炭,搅拌脱色10min。过滤,滤液用氯化氢乙醇溶液中和,冷冻结晶,过滤,用丙酮洗涤结晶,70℃以下干燥,即得古柯碱盐酸盐。

用途:具有局部麻醉作用,其0.03%水溶液即能麻醉感觉神经未梢。古柯碱及其盐酸盐的水溶液在煮沸消毒时分解,应用不方便,现多被其他合成的局部麻醉药所代替。目前,眼科仍常用其作粘膜麻醉剂。古柯碱对中枢神经有较大的毒性,能使大脑皮层兴奋产生欣快症,反复使用,可迅速产生柯瘾,成为一种毒品。
