Based on the results of genotype identification,the waxy wheat s improved BC5F2 backcross progenies were used in this study to verify the effects of null alleles on reducing amylose content,determine the amylose synthesis capacity of each Wx gene,and investigate the correlation of amylose content with agonomic traits,quality traits and starch pasting properties,respectively.

In order to produce selectable marker-free (SMF) transgenic rice containing antisense Wx gene, two T-DNA regions, with the HPT selectable marker gene and the antisense Wx gene, respectively, in two binary vectors or a super-binary vector, were constructed and co-introduced into several rice cultivars, both of Japonica and Indica, by Agrobacterium tumeficien(s)-mediated co-transformation system.
本试验以具不同直链淀粉含量的粳稻品种粳9983和广陵香粳,籼型杂交稻重点亲本协青早、龙特甫和珍汕97为受体材料,经根癌农杆菌介导的双载体或超双元载体共转化技术将分别含有潮霉素抗性(HPT)选择标记和反义Wx基因的两个T-DNA区导入上述受体来源的愈伤组织中,通过检测T_0代转基因水稻植株中HPT基因与反义Wx基因的共整合情况及在T_1代植株间的分离情况,来研究不同的共转化技术路线中共转化的效率及对去除抗性选择标记的效果,进一步为用基因工程技术改良稻米品质提供依据和育种材料。
