To get the partial coding region of the duck GHSR mRNA and to detect the mutation loci in the cloned sequence,RT-PCR was carried out to isolate the target sequence from the hypothalamic tissue of the native breed,Chaohu duck,and then cDNA pooling with 30 cDNA samples was amplified and sequenced for the mutation detection of the cloned sequence.
采用RT-PCR法从巢湖鸭下丘脑组织中分离家鸭GHSR基因mRNA中编码区核酸序列,并选用30个个体cDNA,通过构建cDNA池对克隆编码区的序列变异测序检测。
