性质:以硅胶为固定相的柱色谱法。在经典色谱法中,色谱管为内径均匀、下端缩口的硬质玻璃管,下端用棉花或玻璃纤维塞住,柱管内装入硅胶吸附剂。硅胶的颗粒应尽可能保持均匀,通常直径为:0.07~0.15mm。装柱时将硅胶与流动相相混合,搅拌除去气泡,徐徐倾入色谱柱中。在经典柱色谱中,亦可采用干法装柱。在HPLC中一般采用粒度为5~10fzm的球形或无定形硅胶,柱管长为15~25cm,内径多用0.46cm,采用匀浆法装柱。当采用3恤硅胶时,柱效大于80 000片/m;当采用10μm硅胶时,柱效可大于25 000片/m。它可以进行吸附色谱法的分离,当硅胶含水大于17%时则进行分配色谱法的分离。
