脱氢酶 dehydrogenases 一类催化物质氧化还原反应的酶,在酶学分类中属于第一大类。反应中被氧化的底物叫氢供体或电子供体,被还原的底物叫氢受体或电子受体。当受体是O2时,催化该反应的酶称为氧化酶,其他情况下都称为脱氢酶。不同的脱氢酶几乎都根据其底物的名称命名 。大多数脱氢酶的天然受体是NAD+或NADP+〔以下用NAD(P)+表示〕,例如苹果酸脱氢酶,异柠檬酸脱氢酶等 。脱氢酶的底物经这类脱氢酶的催化使NAD(P)+还原生成NAD(P)H。另一些脱氢酶以黄素为辅基,辅基在催化反应中进行氧化还原。NADH脱氢酶、琥珀酸脱氢酶都含有黄素辅基。NADH脱氢酶、琥珀酸脱氢酶均含有非血红素铁和酸不稳定硫。铁硫中心不稳定,往往是酶失活的原因,铁硫中心也是一些酶抑制剂的作用部位。生物体中绝大多数氧化还原反应都是在脱氢酶及氧化酶的催化下进行。物质经脱氢酶催化氧化,最后通过电子传递链而被氧氧化,此时通过氧化磷酸化作用生成腺苷三磷酸(ATP),是异养生物体取得能量的主要途径。 |
