Objective A simple method for detection of hepatoma-specific gamma-glutamyltranspeptidase(HS-GGT)was set up by Datura stramonium agglutinin(DSA)affinity chromatography,and its clinical application was verified in differential diagnosis of primary hepatocellular carcinoma(PHC).
方法采集64例PHC、22例肝外肿瘤、68例良性肝病及21例正常对照者的血清,以神经胺酸酶(NMD)水解后直接用欧曼陀罗凝集素(DSA)亲和层析柱分离,首先用柱平衡液洗脱不结合组分,再用50mmol/L醋酸洗弱结合组分,最后用100mmol/L醋酸洗脱强结合组分,用连续监测法测定各组分中的GGT活性,并分析各组分GGT在PHC鉴别诊断中的价值。

| 曼陀罗中毒 daturism 误食茄科曼陀罗属植物的种子、浆果或幼苗而引起的有毒植物食物中毒，症状以神经系统异常为主。 曼陀罗多生长于田间、沟旁、道边、河岸等处，其全株均有毒，毒性物质为茛菪碱、东茛菪碱和阿托品等。有报道误食其果实3粒即中毒者。 中毒的临床表现类似阿托品中毒。潜伏期短者20分钟，长者3小时，多为1小时左右。症状先有唇干口渴、噪子发苦、吞咽困难 、声音嘶哑、皮肤干燥潮红 、发热 ，继而有头晕、瞳孔散大、视力模糊，甚至幻听幻视，严重者心率加快、躁动不安、血压不稳、惊厥、昏迷，甚至因呼吸衰竭而死亡。 急救措施为催吐和洗胃，迅速清除毒物，减少体内吸收；解毒治疗可皮下注射毛果芸香碱，也可皮下注射水杨酸毒扁豆碱，对症治疗主要是针对神经系统症状和呼吸系统症状。 预防措施主要是加强宣传教育，尤其是教育儿童识别并不吃曼陀罗的浆果，防止曼陀罗的种子混入可食豆类，防止曼陀罗的幼苗、叶子混入菠菜等蔬菜中，在食用菠菜等蔬菜时，要挑除菜叶的杂草杂叶。 |
