性质:3′,5′-环鸟苷酸。C10H12N5O7PMr345.2。在鸟苷酸环化酶(GC)催化下由GTP合成;在CGMP磷酸二酯酶(PDE)催化下水解。主要在心肌、血管平滑肌、小肠上皮、脑、肾集合管细胞中起第二信使作用,其效应因组织而异,如利钠肽激活质膜GC,在血管使平滑肌舒张、血压下降,在肾使Na+、水排出增加,血容量降低。心肌和血管平滑肌细胞中还存在另一种GC固工酶,是一种与血红素紧密结合的胞浆蛋白,其天然配体是一氧化氮。在心脏中,当NO与GC的血红素基结合后,GC活性增加,CGMP合成增加,刺激保持低胞浆[Ca2+]的离子泵,降低心肌收缩力。硝酸甘油类药物治疗心绞痛的机制即在于能自发分解产生NO。CGMP的大多数作用均由CGMP依赖的蛋白激酶(G激酶)中介。但在视杆细胞外节圆盘膜接受光照时,视紫红质变构,通过G蛋白,激活PDE,调节CGMP水平而引起Na+通道一时性关闭,最终发生超极化现象。
