性质: 肼在生物化学中,多肽与无水肼加热而发生反应,除羧基末端氨基酸以游离形式存在外,其他的氨基酸都转变为相应的氨基酸酰肼化合物的反应。在蛋白质肽链一级结构测定中确定羧基末端氨基酸的一个最重要的化学方法。反应中生成的氨基酸酰肼可与苯甲醛作用变为水不溶性的二苯基衍生物沉淀,上清液中的游离羧基末端氨基酸借助FDNB(二硝基氟苯)法或DNS(丹磺酰氯)法以及层析技术进行鉴定。肼解过程中,谷氨酰胺、天冬酰胺、半胱氨酸等被破坏,故不易检测。羧基末端如果是精氨酸,则将转变为鸟氨酸。用化学方法测定脱氧核糖核酸序列过程中,用肼使嘧啶环断开的反应也同样属于肼解的结果。
