诊法。免疫标记技术之 一。用于测定抗精子抗体。原理为将欲测血清与相应的固相抗原形成免疫复合物，由标记上 酶(过氧化氢酶、碱性磷酸酶、尿素酶等)的二抗检测。利用这结合的酶使基质(邻苯二胺、 对硝基苯酚、尿素等)转化为色素原，出现的颜色反应以目测或分光光度计测出。方法：用 PBS洗涤精子，将其密度调至25×106/50μl。取50μl加到微量滴定盘的每一凹孔中， 以025%戍二醛固定。用含005%Tween20(表面活性剂)的PBS(PBS-Tween)洗3次，加入50 μl稀释的血清或精浆(1∶4稀释)，在37℃下孵育60分钟后，用PBS-Tween液将凹孔洗3次， 加入50μl，1∶100稀释的联结有磷性磷酸酶的羊抗人免疫球蛋白，经37℃60分钟孵育，再 用PBS-Tween液洗3次，加入250μl含1mg/ml磷酸对硝基苯酚的005M碳酸钠缓冲液，3 7℃60分钟孵育后，每孔加入50μl3NNaOH以中止反应。用分光光度计在400nm测定吸收光 谱。含磷酸对硝基苯酚和3NNaOH作为空白。本法优点在于不需新鲜精子，适宜大量标本测定 ，但与无关抗原的非特异性反应是主要问题。
