本课题采用基因工程重组DNA技术克隆ICOS活性片段、构建ICOS-mlg融合基因真核表达载体,在哺乳动物细胞中实现ICOS-mlg的高效表达,并通过亲和层析技术纯化重组蛋白;在此基础上,进行了ICOS-mlg体内外生物学功能的研究;观察了ICOS-mlg阻断共刺激通路对过敏性哮喘炎症的影响;采用Insightll分子模拟的方法和配体亲和筛选突变点,并构建、表达获得配体高结合活性ICOS突变体融合蛋白,用异基因混合淋巴细胞增殖抑制试验检测突变体蛋白的生物学活性;另外经过反复实验,完成了基因工程人源ICOS-Ig的纯化工艺研究,为下一步中试规模的制备和实验研究打下了基础。

allergic rhinitis
又称“过敏性鼻炎”。为身体对某些变应原敏感性增高而呈现以鼻黏膜病变为主的一种异常反应，常伴过敏性鼻窦炎。临床上有常年性发作和季节性发作二型，我国的前者为多，可发生于任何年龄。临床上多以阵发性鼻痒为先发症状。常见的症状有鼻塞、流清水样鼻涕、鼻及咽部发痒、打喷嚏等、鼻分泌物涂片可见嗜酸性粒细胞。发作期查鼻黏膜苍白水肿。治疗主要有脱敏疗法、抗组胺药或肾上腺皮质激素药物治疗及中医中药治疗和局部治疗；若伴有鼻息肉、鼻甲肥大或鼻中隔偏曲时，应考虑手术治疗。
