卵黄囊由内胚层和包于其外方的胚外中胚层组成。其顶部的内胚层构成原始消化管，其他部分留在胚体之外，称固有卵黄囊与原始消化管借卵黄蒂相连。约在胚胎发育第5周，卵黄囊蒂缩窄闭锁，卵黄囊与原始消化管断离，逐渐退化为一直径不到5mm的小泡，残存于胎盘的胎儿面脐带根部附着处。
