A backcross inbred lines (BILs) population consisting of 98 lines derived from a cross of Nipponbare (japonica)/Kasalath (indica)Nipponbare was used to detect QTLs controlling the lysine content of milled rice in two distinct environments,Zhejiang and Hainan Provinces, China.

Using normal maize and o2 maize as materials,the four colorimetric methods of ninhydrin,acid orange-12 dye binding lysine(DBL),2-Chloro-3,5-dinitropyridine(CDNP) and 2,4,6-Trinitrobenzene sulfonic acid(TNBS) were compared and discussed in the regression equation of calibration curve,the lysine content determined by means of these methods,the assay principles and the laboratorial work.

The results indicated: (1 )The amylose content of Hanyou 195 rice was lower than the content of Hanyou 1027 rice;and the relative content of lysine of the Hanyou 195 was hisher than the content of Hanyou 1027 rice.
