由胃黏膜腺体所分泌的黏液覆盖在黏膜表面，形成保护层。它除有润滑和保护胃黏膜免遭食物的机械性损伤作用外，还能有效地阻挡胃腔内的H+接触胃黏膜。胃黏膜上皮细胞顶部的细胞膜相邻连接致密，构成又一道屏障。由于细胞膜为脂蛋白，因此，非脂溶性物质很难透过黏膜层，从而阻碍了胃腔内的H+大量逆向黏膜内扩散，胃黏膜上皮细胞分泌的HCO3-，从黏液深部向表面扩散，遇有由胃腔黏液表面向深部扩散的H+时，即发生中和，使黏液层内形成pH梯度，黏液层腔面pH值低，而黏液层深部的pH值呈中性或偏碱，使胃蛋白酶缺乏起作用所需要的pH环境，从而防止了胃酸和胃蛋白酶对黏膜的损害。
