Methods: Cytotoxicity of diosgenin on U-2OS cell line was determined with MTT assay, its effect on the mRNA expression of matrix metalloproteinase 9 (MMP-9) and tissue inhibitors of matrix metalloproteinases 1 (TIMP-1) in U-2OS cell line was investigated by semi-quantitative RT-PCR analysis.
为探讨薯蓣皂苷元抗肿瘤作用,采用四氮唑盐还原法(MTT法)观察薯蓣皂苷元对人骨肉瘸U-2OS 细胞体外生长的作用,运用RT-PCR方法探讨人骨肉瘤U-2OS细胞在薯蓣皂苷元作用后,其基质金属蛋白酶- 9(MMP-9)和基质金属酶组织抑制剂-1(TIMP-1)mRNA所发生的变化,结果薯蓣皂苷元对U-2OS细胞的生长具有明显的抑制作用,IC50(24h)为45 μmol／L:薯蓣皂苷元抑制U-2OS细胞MMP-9基因转录,而TIMP-1基因转录不受影响。
