体内试验发现，由缺乏终止密码子的mrna所编码的λci阻遏蛋白和细胞色素b562的c-末端也连有标记肽，而且标记肽对c-末端的附着依赖于编码tmrna的ssra基因的存在。据此可推测，当由无终止密码子的mrna编码的多肽被翻译时，tmrna才能编码生成标记肽。keiler等据此提出了肽合成的反式翻译模型。反式翻译模型提出，当翻译停止在无终止密码子的mrna的3′-末端时，负载着ala的tmrna进入核糖体(a位)。tmrna将其ala提供给合成停止的多肽链，通过移位作用，核糖体从该mrna进入tmrna orf25编码标记肽的编码区，翻译合成标记肽，它通过ala连在先前合成的肽的c-末端，形成嵌合蛋白。最后，该嵌合蛋白利用框内终止密码子从核糖体上释放出来，使得核糖体进入再循环。tmrna在整个反应中承担着双重功能，先是作为trnaala，接着又作为编码标记肽的mrna。
