制备方法:肝素最初从肝脏中提出,现多由猪、羊、牛等动物的肠粘膜中提取,从肺脏,肝脏出可提取。取新鲜肠粘膜投入反应锅内,按3%加入氯化钠,用40%的氢氧化钠调节pH至9.5。在半小时内逐步升温至60-65℃,继续保温搅拌2h,然后升温至95℃,维持10min,冷却至50℃以下过滤。将强碱性季铵盐阴离子树脂加入滤液中,搅拌吸附8小时,静置过液。次日,虹吸去上层液,树脂用水冲洗至上清液澄清,滤干,用2倍量1.4M氯化钠搅拌2小时,滤干,树脂再用不着倍量1.4M氯化钠洗涤1次。然后,树脂用3M氯化钠洗脱两次,滤干,将滤液合并,加入等量的95%乙醇沉淀过液,收集沉淀,用丙酮脱水干燥,得粗制品。粗制品溶于1%氯化钠中,用盐酸调节pHGCF 1.7-1.8,过滤。用5N氢氧化钠调节pH至11,加30%浓度过氧化氢脱色,过滤。滤液用盐酸调节pH至6,加入等量95%乙醇沉淀。沉啶物用丙酮洗涤,干燥,即为肝素。
用途:该品是制抗凝药肝素剂的原料。
