Objective To investigate the role of Neurogenic mechanism and the mechanism of cardio-protection of medico-intervene during acute myocardial ischemia the observation was made on the changes in serotonin(5-HT) in the ischemic and the non-ischemic myocardium,dorsal root ganglion,comu dorsale medulla spinalis and comu rentrale medulla spinalis induced by coronary artery occlusion (CAO) in rats.
5h)、T2(1h)、T3(3h)和T4(6h)四个时点,分别采用免疫组化和原位分子杂交技术观察冠脉结扎大鼠不同时间血运阻断区和非血运阻断区心肌、背根神经节以及脊髓背角和腹角内5-HT的变化;在第一部分实验的基础上选用270-300g雄性SD大鼠随机分为六组:对照组(冠脉下穿线不结扎)、单纯缺血组(结扎冠脉)、吗啡组、立其丁组、艾司洛尔组、阿托品组,吗啡、立其丁、艾司洛尔、阿托品干预组在CAO前15min分别经尾静脉注射吗啡1。

方法:第一部分实验选用270-300g 雄性SD 大鼠随机分为四组:对照组(C 组)、扎闭冠脉1h 组(CAO1h 组)、扎闭冠脉3h 组(CAO3h 组)和扎闭冠脉6h 组(CAO6h 组),分别采用免疫组化和RT-PCR 技术观察冠脉结扎大鼠不同时间血运阻断区和非血运阻断区心肌以及背根神经节内SP 和CGRP 的变化;在第一部分实验的基础上选用270-300g 雄性SD 大鼠随机分为四组:对照组(冠脉下穿线不结扎)、单纯缺血组(结扎冠脉)、吗啡干预组和曲马多干预组。
