该液用于体外培养,1000ml超纯水或去离子水经重蒸后含Ham’s F10干粉9.8g,硫酸镁(MgSO4·7H2O)0.124g(1mM),青霉素G(K)(penicillin G(K))0.05g,乳酸钙(calcium lactate·3H2O)0.3g(1mM),碳酸氢钠(NaHCO3)1.68g(20mM),碳酸氢钾(KHCO3)0.50g(5mM)经过滤分装,渗透压280~290 mOsm/kg H2O pH 7.35~7.45可在4℃下保存,2周内用完。生长液中含灭活人血清15%;授精液中含10%。
