螺旋体血细胞凝集试验


  诊法。梅毒实验 室诊断方法之一。原理:抗原是把Nichols株螺旋体用超声波打碎成可溶性抗原,再与已 经福尔马林固定,用鞣酸处理的绵羊红细胞混合,以羊血细胞为载体并吸附螺旋体可溶性 抗原而成。被检测的血清标本是用Reiter株非致病性螺旋体经半固定培养基繁殖一定数量后 ,高压灭菌及高速离心后的上清液,即为吸收剂。用此吸收剂稀释血清,可将血清中非特异 性 抗体吸收掉,并不影响特异性抗体的滴度。取经吸收剂稀释并吸收过的血清于血凝反应板上 ,每孔25μl,用特制的滴管于已滴加25μl血清的孔内再加入已吸附抗原的羊血细胞3滴( 75μl)将反应板放在微型振荡器上混合5分钟,使致血细胞与吸收过的血清充分混合, 放于25℃4小时或4~8℃冰箱中 过夜即可观察结果。对照非致敏血细胞及阴性、阳性血清,结果明显,容易判定。
