雌激素能使绝经后加速的骨转换率恢复绝经前状态,减少骨丢失,可预防发生绝经后骨质疏松症;绝经后及时应用并持续5年以上者,脊椎骨折率及髋部骨折率均比不用者低。其作用机制未全阐明,但均认为雌激素经多途径影响骨代谢:①降低骨对甲状旁腺激素的敏感性,减慢骨的吸收;②增加体内降钙素量,减少骨吸收;③加快肠道钙吸收;④减低肾对钙的排出;⑤对骨骼有直接作用,通过成骨细胞释放的细胞素,降低破骨细胞的功能,抑制骨吸收。而且雌激素对缓解更年期症状有特效,现多选做预防骨质疏松药物,口服或经皮吸收方便。剂量因不同制剂而异。
