诊法。免 疫标记技术之一。用来测定抗精子抗体。原理为用整个精子,或溶解的精子膜,或人工合成的 精子加肽抗原作为抗原,包裹在直径为3mm的聚苯乙烯珠上,用125I标记的蛋白A(一 种从葡萄球菌中提取的蛋白,具有吸附IgG的能力)加以测出。步骤:用003M磷酸缓冲液(pH72)将血清样品稀释100倍,在试管中与包裹抗 原的小珠在37℃下孵育30分钟,再于25℃下放置30分钟。将孵育液吸弃,用1ml005M巴比妥 缓冲液冲洗两次。125I-蛋白A溶于含001%牛血清蛋白的005M巴比妥缓冲液(pH 9 0),浓度为60000cpm/200μl。在每个试管中加入200μl标记液。然后试管在3 7℃ 下再孵育30分钟,重复冲洗过程。最后在γ仪上计数1分钟。每个样品重复3次。每一批实验 中包括一个阳性对照血清(兔抗人精子抗血清1∶100,1∶1000,1∶2000,1∶4000和1∶8000)作为质量控制。本法操作简便,适于做大量样品。
