性质: 又称色谱聚焦。它是根据蛋白质等电点,结合离子交换技术的大容量色谱。其原理是利用离子交换剂本身的带电基团的缓冲作用,当洗脱缓冲液滴到离子交换剂上,可自动形成pH值梯度,而被分离的蛋白质其所带的电荷及其移动行为取决于它们的等电点和介质的pH值。当环境的pH值低于等电点,蛋白质呈正电荷,不与阴离子交换剂结合而向下移动;当环境的pH值高于蛋白质等电点时,蛋白质带负电荷,与阴离子交换剂结合。随着洗脱过程的进行,所形成的pH值梯度也不断下降,蛋白质与阴离子交换剂的结合和脱离过程不断重复,但当一种蛋白质在柱上随洗脱液下移至等电点处,此时其移动速度明显减慢,并且相同的蛋白质会聚焦在一起,按等电点顺序从柱底流出。
