性质:是一种将DNA分子在体外诱发位点专一突变的技术。常用方法有(1)D环诱变法。将同源单链DNA小片段与完整超螺旋双链DNA分子混合在含有ReCA蛋白,ATP的缓冲液中,由于单链侵入双链而在同源区域产生一个单链D环。用单链DNA专一性诱变剂二亚硫酸盐处理上述DNA,可使D环上胞嘧啶脱氨而成为尿嘧啶。将处理过的DNA转化ung-细胞,经过DNA复制,尿嘧啶与腺嘌呤配对,导致G·C到A·T的转换。(2)寡核苷酸诱变法。将一个化学合成的含有突变碱基顺序的寡核苷酸片段与单链靶DNA分子退火复性形成异源双链核酸分子,由该寡核苷酸片段作为引物,通过Klenow酶催化合成第二条链。在连接后,将该双链DNA分子(可自主复制)转化到宿主菌,经复制使突变链和非突变链分离,能获得含有预期突变的细胞。这个技术与传统诱变剂方法相比,具有突变率高、突变位点和碱基顺序明确等优点。
