性质:也有人称作上皮生长因子。是一种低分子量,对热稳定和不可透析的多肽。能直接刺激表皮的生长和角化。其作用如下。(1)促进细胞增殖:能促进鼠和人的成纤维细胞、神经胶质细胞、乳腺上皮细胞、角质细胞、心血管内皮细胞等的增殖;还促进HeLa细胞及SV40-3T3细胞等传代细胞的增殖;但也有人报道能抑制人表皮样细胞瘤细胞A431的生长;还能促进细胞中DNA,RNA,蛋白质以及细胞外大分子的生物合成。(2)增加物质的转运和糖酵解:EGF能通过快速增加乳酸产量来激活静止期3T3细胞的糖酵解。EGF的作用是经Ca2+介导的。(3)对花生四烯酸的释放及前列腺素的合成的影响:EGF能激活磷酯酶A2,从而促进上皮细胞膜释放花生四烯酸(AA),无外源性Ca2+的条件下,生理浓度的EGF(1ng/ml~0.1μg/ml)即能刺激AA的释放。加人Ca2+后(0.3~1.2mmo1/L)能显著加强此种效应。(4)对磷酯酰肌醇代谢的影响:能促进磷酯酰肌醇的代谢和钙的内流(EGF对磷酯酰肌醇代谢的促进作用明显依赖于钙的内流),能刺激第二信使三磷酸肌醇和甘油二酯的生产。能激活钙和磷酯依赖性激活酶(蛋白激酶C)而且还能刺激磷酯酶A2之活性和增加前列腺素的释放。与尿抑胃素为同一物质。从小鼠颌下腺分离或基因工程技术制得的一种细胞因子。另外在体内介导上皮生长,促进血管形成,抑制胃酸分泌;加速伤口愈合。医疗上用于角膜与晶体手术、伤口愈合、皮肤溃疡、慢性软组织溃疡的治疗。
