In order to search for more potent and less toxic antithrombotic agents, eleven derivatives of 6-(4-substituted-phenyl)-4, 5-dihydro~3(2H)-pyridazinones were designedand according to mechanism of action and structure-activity relationship of dihydropyridazinone compounds.
为了寻找优良的血小板聚集抑制剂,本课题依据二氢哒嗪酮类化合物的作用机理和构效关系,在保留二氢哒嗪酮类化合物基本结构的基础上适当延长碳链,引入各种不同的侧链及杂环,共设计并合成了十一个二氢哒嗪酮类化合物,均为首次报道。

In order to search for more potent and less toxic antithrombotic agents twenty-eight derivatives of 6-(4-substituted aminophenyl)-5-methyl-4,5-dihydro-3(2H)-pyridazinones and 6-(4- substituted aminophenyl)-4,5-dihydro-3(2H)-pyridazinoneswere designed and synthesized according to mechanism of action and structure-activity relationship of dihydropyridazinone compounds.
二氢哒嗪酮类化合物具有良好的心血管药理作用如抗血小板凝集和强心等,有的已成为药物如匹莫苯(pimobendan),有的已结束Ⅲ期临床研究如MCI-154,有些正在进行临床前研究如SMⅡ_4、Y909、DMDP和CCI-17810等,这类化合物在心脑血管疾病的治疗方面具有广阔的应用前景。
