为探讨SLA及转铁蛋白多态性与仔猪大肠杆菌K88腹泻的关系,本研究利用PCR产物直接测序技术、PCR-SSCP技术和不连续聚丙烯酰胺凝胶蛋白电泳技术,以两个中国地方猪种久仰香猪、剑白香猪和一个外国猪种长白猪同胞半同胞0-28日龄仔猪混合家系试验猪群为试验材料,研究了SLA-DQB外显子2、SLA-DRB外显子2、SLA-DQB近端调控区、SLA-DRB近端调控区以及转铁蛋白的多态性,分析了这些多态位点对0-28日龄仔猪大肠杆菌K88ab、K88ac和K88ad小肠粘附型以及血清IgG含量的影响,以寻找有用的遗传标记应用于猪的抗病育种实践。

The exon 2 of SLA(swine leukocyte antigen)-DQB and SLA-DRB genes in Sutai pigs was analyzed by PCR-RFLP method,and the results showed that the exon 2 of SLA-DQB gene digested by RsaⅠcould be divided into three kinds of alleles and five kinds of genotypes,the exon 2 of SLA-DRB gene digested by RsaⅠcould be divided into three kinds of alleles and four kinds of genotypes.
结果表明,苏太猪SLA-DQB基因经RsaⅠ酶切后共产生3种等位基因,5种基因型;SLA-DRB基因经RsaⅠ酶切后共产生3种等位基因,4种基因型。

The FUT1 (Escherichia coil F18 receptor-related gene, ETECF 18 gene) and SLA-DQB gene were identified as candidate genes for disease resistance in pig, which were very important in the research field of pig breeding for disease resistance.
大肠杆菌F18受体(ETEC F18R)相关基因(FUT1)和SLA-DQB基因是与猪抗病育种相关的重要候选基因,本研究分析了猪大肠杆菌F18受体(ETEC F18R)相关基因(FUT1)和SLA-DQB基因多态性及其与生产性能之间的关系,进一步探索了外来猪种与中国地方猪种间FUT1基因和SLA-DQB基因遗传基础的差异,重点探讨了不同基因型猪F18抗性/易感性的相关性,旨在建立猪抗F18大肠杆菌病分子育种的可行性方案,主要结果如下: (1)采用PCR-RFLP和PCR-SSCP技术,分别对14个猪种FUT1基因开放阅读框307bp和857bp位点进行检测,M307位点只有在外来猪种杜洛克、皮特兰中检测到AA型纯合子,外来猪种中的约克夏、长白猪以及所有的国内地方猪种和含有外来猪种血缘成分的猪种均未发现AA型纯合子,国内地方猪种大部分个体为GG型纯合子,其基因型频率为0。

This study was conducted to analyze the polymorphisms of SLA-DQB gene in native and exotic pig breeds and aimed to provide a theoretical foundation for further research on correlation between SLA-DQB gene and disease resistance.
本研究旨在系统分析国内外猪种SLA-DQB基因的多态性,以期为进一步研究猪SLA-DQB与疾病抗性的相关性提供理论依据。
