Objective:To detect expression of the genes associated with TGF-β/Smad signaling pathways in RA patient and evaluate their effect in RA,evaluate their expressions in progress and outcome of RA patient,to investigate the ralation of those detected factors and the syndrome types of Traditional Chinese Medicine(TCM),to provide an important theory of diagnosis and treatment in RA.
研究目的:建立检测类风湿关节炎(RA)外周血单个核细胞(PBMC)中TGF-β/Smad(转化生长因子-β/Smad)信号转导通路相关因子的表达水平的实时荧光定量PCR的方法,评价TGF-β/Smad信号转导通路相关因子在RA中的表达水平,及其在疾病的发生、发展、归转等方面的临床意义;并探索其在RA中医证候分型中的作用,为研究RA的发病机制及治疗提供新的理论根据。

Methods The influence of tetramethylpyrazine on the growth and proliferation of fibroblasts was detected through the incorporation H3-thymidine into DNA taking NIH3T3 as the model and on the expression of collagen I was observed by immunoluciferase stain test,and on conduction of TGF-β/Smads signal was studied by using the cell line of Mv1Lu-(CAGA)12-Luc.
方法以NIH3T3成纤维细胞为模型,通过H3-thymidine DNA掺入量测定川芎嗪对成纤维细胞生长和增殖的影响;通过Ⅰ型胶原的免疫荧光染色实验,观察川芎嗪对成纤维细胞Ⅰ型胶原蛋白表达的影响;并利用Mv1Lu-(CAGA)12-Luc稳定细胞系,探讨川芎嗪对TGF-β/Smads信号传导通路的影响。
