采用膜片钳细胞贴附式和内面向外式记录方式观察川芎嗪对猪冠状动脉平滑肌细胞大电导钙激活钾通道(large-conductance Ca2+- activated potassium channels,BKCa channels)的作用,分别用蛋白激酶A(protein kinase A,PKA)抑制剂H-89和蛋白激酶G (protein kinase G,PKG)抑制剂KT-5823处理细胞,再观察川芎嗪对BKCa通道作用的变化。

calcium channel blockers
又称“钙拮抗剂”、“慢通道阻滞剂”。指能选择性的阻滞细胞膜上的钙通道,减少Ca2+经钙通道内流的一类药物。主要有二氢吡啶类(如硝苯吡啶、尼莫地平)苯烷胺类(如维拉帕米)和硫氮■类(如硫氮■酮)等。在钙通道蛋白上有此三类药物的高亲和力结合位点。具有扩张血管、降低血压、减弱心肌收缩力和抑制血小板聚集等作用。主要用于治疗冠心病、高血压病、室上性心动过速、脑血管疾病等。不同药物对不同部位组织有相对选择性,如硝苯吡啶扩张冠状血管作用比较显著,而尼莫地平扩张脑血管作用较强。
