A photosensitiser temoporfin was synthesized from 3-hydroxybenzaldehyde by acetylation to give 3-acetoxybenzaldehyde, which was subjected to cyclization with pyrrole to give 5,10,15,20-tetra(m-acetoxyphenyl)porphyrin followed by deacetylation, reduction and selective dehydrogenation with an overall yield of about 10%.
间羟基苯甲醛乙酰化后与吡咯环合制得5,10,15,20-四(间乙酰氧基苯基)卟啉,再经脱乙酰基、还原和选择性氧化制得光敏剂替莫泊芬,总收率约10%。

性质:无色针状结晶。熔点161-162℃(157-158℃)。易溶于丙酮,稍溶于甲醇、乙醇,难溶于氯仿、苯,几乎不溶于水。遇光渐变色,无臭。

制备方法:由对氯苯丁酮酸-4经α-溴化后再与硫代苯甲酰胺缩合而得。

用途:氯苯噻乙酸为一种新的甾体消炎镇压痛药,可用于慢性关节炎、变形性关节症、肩周炎、腰痛、呼吸道炎症的消炎镇痛,也用于外伤及术后,拔牙的消炎镇痛。
