性质:被测组分先通过衍生化反应,转化为衍生化产物,然后再经过色谱柱进行分离。高效液相色谱法中柱前衍生的目的是:通过衍生化反应,使原来不能直接被检测的组分转化为能被所用检测器检测的物质;使被测组分与衍生化试剂有选择地参加反应,而与样品中的其他组分分离;改变被测组分在色谱柱上的出峰次序,使之更有利于分离。柱前衍生的优点是可以不必严格限制衍生化反应条件,可以允许较长的反应时间及使用各种形式的反应器。其缺点是当一个复杂组分样品经过衍生化反应后,有可能产生多种衍生化产物,给色谱分离带来困难。
