Then,1-methyl-2,6-diphenylpiperidin-4-one is synthesized using 1,5-diphenylpenta-1,4-dien-3-one and methylamine,At last,Gewald reaction was utilized to obtain ethyl 2-amino-6-methyl-5,7-diphenyl-4,5,6,7-tetrahydrothieno pyridine-3-carboxylate through the reaction of 1-methyl-2,6-diphenylpiperidin-4-one with ethyl cynoacetate and sulfur in the presence of morpholine.
首先利用醛酮缩合制得二烯酮(1),然后利用1与甲胺溶液作用制得哌啶酮(2),最后利用Gewald反应使2、氰乙酸乙酯与硫磺在吗啡啉催化下,得到未见文献报道的2-氨基四氢吡啶并噻吩衍生物3,产率60%～80%。
