性质:又称定点诱变或定位诱变。采用基因合成技术或取相应的寡核苷酸,用酶法(或化学方法)改变基因中某一点上密码子,再把含有该点突变的基因插入到质粒上,形成重组质粒,经转化把重组质粒引入到酵母菌、大肠杆菌等新宿主细胞中进行表达,其表达的产物是一个突变蛋白或突变酶。采用定点突变的方法,可灵活而巧妙地勾画甚至确定某特定氨基酸残基在蛋白质和酶分子中的功能和作用。也是创建工程化蛋白或酶的工具方法之一。
