凯库勒在提出了多种有关苯的开链式结构而又因其与实验结果不符被一一否定之后，1865年，他终于悟出闭合链的形式是解决苯分子结构的关键，他以苯的（ⅰ）式表示这一结构。1866年他又提出苯分子是一个由6个碳原子以单、双键相互交替结合而成的环状链（ⅱ）式，后简化为（ⅲ）式，也就是我们现在所说的凯库勒式。
