质子自旋量子数I为1/2时，其磁矩μN在静磁场H中只有两种取向,分别对应于能量为E-┩＝γ啚H/2及E┩＝-γ啚H/2 的能级(γ为回磁比；啚＝h/2π,h为普朗克常数)。如垂直于H的射频电磁波的频率v 满足hv＝E-┩-E┩＝γ啚H时，则低能级的质子就从射频电磁波吸收能量,跃迁至高能级。将射频电磁波的能量变化对磁场作图，即得质子磁共振谱。
