The plasmid p387NIT1 was transformed into nitratase deficient and cell wall-less strain CC-2677(cw15nit1-305mt-) of Chlamydomonas reinhardtii by using glass bead method, and some insertion mutatedtransformants with various background have been obtained.
微玻璃珠研磨法将p387NIT1质粒转入到莱茵衣藻硝酸还原酶缺陷型及缺壁突变株品系CC-2677(cw15nit1-305mt-)中,得到了一些不同的插入型实变转化子,在本实验室条件下CC-2677突变株品系的转化率最高时达到了88个/μgDNA,经多次转化共获得各种转化子1800多个,对它们进行荧光检测后检测出缺乏叶绿素b的一个突变子NIT17-1,并进一步用高效液相色谱法对其光合色素含量进行检测,结果证实该突变体的确丧失了叶绿素b合成能力,该突变株将可作为CBN1基因克隆及其定位的有利工具。
