2003年夏季利用开苞导入法经花粉管通道,将抗旱耐盐基因HVA1和碱蓬、芦苇的DNA导入到稳定的玉米自交系中,获得2096粒种子,2003年冬至2004年夏,通过苗期反复干旱法及田间标记性状抗旱鉴定,对后代材料进行了详细的观察和分析。

Total RNA in leaves of Poa pratensis cultivars under drought stress was extracted for reversing transcription into cDNA and then the cDNA was as a template for PCR reaction according to designing primer of cds of Hordeum valgare HVA1 drought resistance gene(X78205).
根据大麦(Hordeum valgare)HVA1抗旱基因(X78205)的mRNA序列设计引物,提取干旱胁迫下草地早熟禾叶片中的RNA,以反转录后的cDNA为模板,进行PCR反应,扩增产物经回收、连接、转化和酶切鉴定为阳性重组子,其序列长度为324bp。
