To develop the genetic markers linked to broodiness,the 5′region of the goose prolactin receptor(gPRLR) gene from a matured male Hongzehu Goose testis tissue was defined using RACE.

The gel was electrotransferred on to ni-trocellulose membrane, combined with prolactin receptor antibody and HRP-conjugated secondary antibody, acted on luminol chemiluminscent substrate,exposed against X--ray film, developed.

Using rapid amplification of cDNA ends(5′-RACE),three 5′-end fragments of goose prolactin receptor(gPRLR) cDNA of 361 bp,499 bp and 594 bp in adult male goose testis were isolated.

A DNA fragment of Peking duck prolactin receptor(PRLR)spanning 1 325 bp was obtained by PCR method.
