To provide a basis for further investigation of the chicken interferon-γ(ChIFN-γ),a pair of primers specific to ChIFN-γ were designed and synthesized according to the known sequences from GenBank,and were used to amplify ChIFN-γ cDNA by RT-PCR from total RNA of chicken spleen cells induced by ConA.
为了研究鸡γ-干扰素(ChIFN-γ)基因的功能并进一步探究其在控制家禽传染病方面的应用,根据GenBank已发表的ChIFN-γcDNA基因序列自行设计引物,应用反转录-聚合酶链式反应(RT-PCR)技术从伴刀豆球蛋白A(Co-nA)刺激培养鸡脾淋巴细胞中提取的总RNA中扩增得到ChIFN-γ,然后连接到pMD18-T载体上并转化到Top10感受态细胞中,获得阳性重组质粒并进行正确性序列测序。
