一种体外测定肿瘤病人细胞免疫状态的方法。将肿瘤组织消化处理，使细胞分散，然后接种800～1000个瘤细胞于直径6厘米的平碟中培养，使成单层细胞培养，可得到20～l00个细胞集落。将待测机体的淋巴细胞加入到培养的肿瘤集落中，培养3～5天；染色计数细胞集落数。对照加入正常人淋巴细胞代替待测的淋巴细胞。比较肿瘤病人和正常人淋巴细胞抑制率或治疗前后淋巴细胞免疫功能的变化情况。淋巴细胞免疫功能强者对癌细胞有一定的破坏作用。用此法亦可测定血清中有无封闭因子。封闭因子能阻止细胞对癌细胞的破坏作用，癌细胞集落生长不受影响或少受影响。
