      测定质点在重力场中的沉降速度以计算质点大小的分析方法,只适用于粗分散体系。对于质点很小的分散体系(<0.1微米),由于质点的扩散作用,重力沉降法已无法应用。但如果把分散体系置于强离心力场中(如用超离心机),则质点的大小仍可用沉降方法来测定。用超离心机测定质点大小时,常用的有沉降平衡和沉降速度两种方法。
  
  沉降平衡法 此法采用的离心力场约为重力场的104倍。质点在离心力的作用下向沉降池的底部移动,因而形成浓度梯度。体系中一出现浓度梯度,则发生扩散,而且方向与沉降相反。当扩散与沉降达到平衡后,沉降池中各处的浓度不再随时间而变。这时可根据下式计算单个质点的质量m或高分子的分子量Μ:

  式中k和R分别为玻耳兹曼常数和气体常数;T为热力学温度;v为质点的偏微比容;ρ为介质的密度;w 是转动角速度;x是沉降池中某一位置离转轴的距离;c1和c2是x1和x2处的浓度。因此,只要在沉降达到平衡后测定沉降池中不同位置的浓度,即可算出质点的大小。
  
  沉降速度法 若离心力场强度增至重力场的105~106倍,则扩散作用可以忽略,质点在离心力的作用下运动。当质点所受净的离心力与粘滞阻力达到平衡时,从理论上可导出:

  式中x1和x2为时间t1和t2时质点在沉降池中位置,实际上也是介质与胶体体系的界面在t1和t2时的位置。因此,只要测定不同时间界面移动的位置即可求出 S。但要求出质点的大小则还需要体系扩散的数据。在推导式 (3)和 (4)的过程中,未考虑质点移动时的互相干扰。因此,两式中的D和S均应是无限稀释时的扩散系数和沉降系数。为此,要求出D、S与浓度的关系,然后通过外推方法求出浓度趋近于零时的D和S。
