To investigate the protective effect of astrocyte-conditioned medium (ACM)on anoxia damaged neurons, ACM was collected after cortical astrocytes from neonatal KM mouse isolated, purified and passaged three times, and added into medium of anoxia damaged neurons.
为了探讨星形胶质细胞条件培养液(ACM)对缺氧损伤神经元的保护作用,本实验将新生的KM小鼠的皮层星形胶质细胞分离、纯化并传代培养第三代第5d后,将星形胶质细胞(Ast)条件培养液,加入到缺氧损伤的神经元细胞培养液中,观测其对缺氧神经元的存活及其合成和释放一氧化氮(NO)、乳酸脱氢酶(LDH)和胞膜ATP酶(ATPase)的影响。

方法首先建立hMSC的分离培养扩增体系,制备小鼠胎肝基质细胞条件培养液(FLSC-CM),将hMSC接种在分别含FLSC-CM和IL-6、SCF组合的培养体系中,通过形态学、直接免疫荧光染色检测法、粒-单/巨噬系集落形成细胞培养(CFU-GM)对分化细胞进行鉴定。
