BACKGROUND &OBJECTIVE: It is considered that tumor suppressor gene BRCA1 is an important factor in the regulation of cell cycle checkpoint, but the molecular mechanism by which BRCA1 regulates cell cycle G2/M arrest is less known.
方法:应用转染、流式细胞仪细胞筛选(分选)、Westernblot检测分析BRCA1诱导Gadd45的表达;利用CAT酶活性测定的方法检测外源BRCA1对Gadd45启动子所起作用;以流式细胞周期分析方法检测反义Gadd45转染对BRCA1诱导的细胞G2/M期阻滞的影响;采用细胞生长集落形成分析反义Gadd45转染HeLa、HCT116细胞在BRCA1诱导的细胞生长抑制过程所起的作用。
