性质:又称组织型纤维蛋白溶酶原激活剂。天然物为单链分子,含527个氨基酸组成的糖蛋白。受纤溶酶或胰蛋白酶催化水解Arg275~Ile276肽后转变为由单二硫键相连的双链,其氨基端为重链(A链),羧基端为轻链(B链)。轻链与胰蛋白酶等丝氨酸蛋白酶有很高的同源性,含有t-PA的活性中心His322,Asp371,Ser478。t-PA存在于人体很多组织和体液中溶解血纤维蛋白的丝氨酸蛋白(水解)酶。可选择性地高效激活血栓中纤溶酶原成为纤溶酶,后者水解血栓中的纤维蛋白。目前主要从人黑素瘤细胞培养物中提取t-PA,产品有单链和双链,分子量前者为67 000~68 000,后者两链分别为30 000和40 000。t-PA基因工程和蛋白质工程的目标是生产高效的药用t-PA产品。药用t-PA能高效地预防和治疗急性心肌梗塞等血栓性疾病。
